However, if the incorrect label size issue is resolved by carrying out the steps mentioned in your post “Fix print assist” then this may also resolve the barcode issue.

Thanks for the fast reply.
You were right and re-installing print assist fixed the issue. (as I said on the other post)
However, it did not fix the barcode issue.
We followed RM's instructions for the printer settings where the size of the label needs to be set as 3.75 x 6 inches.
This is what's causing the problem. I've changed the settings to 3.99 x 6 inches and although the label is now slightly offset to the left, barcodes are now being printed correctly and readable.
